Authorities have not yet released detailed information about all the fatalities, and the circumstances surrounding each death remain under review.
Video footage recorded on Thursday showed large groups of migrants, primarily Moroccan nationals, walking around the breakwaters at Tarajal Beach before entering local roads in Ceuta.
While many of those crossing appeared to be young men, families with women and small children were also seen among the crowds. Some migrants shouted, "Viva España!" as they entered the Spanish territory, according to a freelance photographer working for the Associated Press.
Officials have not yet confirmed what triggered the sudden increase in crossings into Ceuta.
The Spanish government is expected to assess the situation further during Friday's visit by Prime Minister Pedro Sánchez and Interior Minister Fernando Grande-Marlaska, while humanitarian assistance continues for those already in the enclave.
